Targeting the RB pathway in tumor cells

Abstract

Retinoblastoma protein (RB) is a negative regulator of the cell cycle. In response to environmental stress, p38 MAPK phosphorylates RB in the N-terminal region, increasing the affinity of RB for E2F, repressing gene expression and arresting the cell cycle. Expression of a p38-phosphomimetic RB mutant decreases the proliferation of cancer cells and tumors in mice. In this thesis, we have designed an assay that monitors RB activity in cells to identify compounds that mimic the effect of RB phosphorylation by p38. This assay has been optimized for High Throughput Screening (HTS) and we have tested over 9,000 different compounds. We identified 32 active compounds (hits) that were classified according to their mechanism of action through secondary tests. Our data indicated that 11 drugs inhibit tumor cell growth and 7 of them arrest the cell cycle in G1. Of these 7 compounds, one activated p38, another is a CDK4 inhibitor and 2 increased the affinity of E2F-RB. We have also identified a smaller RBN-E2F1 interaction region, which is helping us characterize the impact of hits with greater precision. We are currently conducting an HTS assay with larger, newer chemical libraries. The compounds identified in this thesis offer a promising antitumor therapeutic strategy applicable to most types of cancers.

La proteína de Retinoblastoma (RB) es un regulador negativo del ciclo celular. En respuesta al estrés ambiental, p38 MAPK fosforila a RB en la región N-terminal, lo que aumenta la afinidad de RB por E2F, reprimiendo la expresión génica y deteniendo el ciclo celular. La expresión de un mutante RB p38-fosfomimético disminuye la proliferación de células cancerosas y tumores en ratones. En esta tesis, hemos diseñado un ensayo que monitoriza la actividad de RB en células para identificar compuestos que mimetizan el efecto de la fosforilación de RB por p38. Este ensayo se ha optimizado para cribado de alto rendimiento (HTS) y hemos analizado más de 9.000 compuestos diferentes. Identificamos 32 compuestos activos (hits) que se clasificaron según su mecanismo de acción mediante ensayos secundarios. Nuestros datos indicaron que 11 fármacos inhiben el crecimiento de células tumorales y 7 de ellos detienen el ciclo celular en G1. De estos 7 compuestos, uno activó p38, otro es un inhibidor de la CDK4 y 2 aumentaron la afinidad de E2F-RB. También hemos identificado una región de interacción RBN-E2F1 más pequeña, lo que nos está ayudando a caracterizar el impacto de los hits con mayor precisión. Actualmente, estamos realizando un ensayo HTS con bibliotecas químicas más grandes y novedosas. Los compuestos identificados en esta tesis ofrecen una prometedora estrategia terapéutica antitumoral aplicable a la mayoría de los tipos de cánceres.
