dc.description.abstractIdentification through forensic methods is crucial in crime investigations. The Polish Genetic Database of Victims of Totalitarianism (PBGOT) is allowing the positive identification of many victims of Communist and Nazi regimes which have remained unknown for a long time. Forensic science is advancing really fast and the application of Next-Generation Sequencing (NGS) techniques in the identification of victims has completely changed the procedures making them faster, more accurate and less propitious to contamination and manipulation errors. The main objective of this project is to present real-life cases of victims identified with the PBGOT as we continue to identify all the remaining victims. Bones such as teeth and femur were chosen for the analysis because DNA is better conserved in long bones and teeth. Samples were cleaned manually and chemically. Afterwards, bone powder was obtained using a freezer mill and it was kept at -20ÂșC. DNA was extracted with PrepFiler BTA Forensic DNA Extraction Kit (Applied Biosystems) and analyzed with GenoProof 3.0.7. The identified victims presented here are Petr B., Tadeusz Stabrowski and Hieronim Dekutowski.ca
