Abstract

The local translation of messenger RNAs (mRNAs) within different neuronal compartments is a prerequisite the formation of long-term memories. mRNAs are localized via active transport along microtubules, however, the components forming mRNA transport complexes are mostly unknown. A recently developed protein interaction screen suggested novel interactions between microtubule motor and RNA-binding proteins. In this thesis, we (1) expressed and purified the identified protein interactors, (2) developed a suitable fluorescence microscopy-based in vitro reconstitution approach, and (3) used it to study the proposed interactions. This enabled us to identify and a novel RNA transport complex that suggests a connection between the nuclear export and active transport of neuronal mRNAs along microtubules. Since one of the factors also functions as a 3’ untranslated region (3’UTR) length regulator, a recently identified determinant of mRNA localization, this thesis also establishes the groundwork for studying and understanding 3’UTR length-dependent mRNA sorting in neurons.
La traducció local d'ARN missatgers (ARNm) dins de diferents compartiments neuronals és un requisit previ per a la formació de memòria a llarg termini. Els ARNm es localitzen mitjançant el transport actiu al llarg dels microtúbuls, però els components que formen els complexos de transport d'ARNm són majoritàriament desconeguts. Un cribratge d'interacció de proteïnes desenvolupat recentment va suggerir noves interaccions entre els motors dels microtúbuls i les proteïnes d'unió a l'ARN. En aquesta tesi, (1) vam expressar i purificar les proteïnes que vam identificar com a interactors, (2) vam desenvolupar un enfocament adequat de reconstitució in vitro basat en microscòpia de fluorescència i (3) el vam utilitzar per estudiar les interaccions proposades. Això ens va permetre identificar un nou complex de transport d'ARN que suggereix una connexió entre l'exportació nuclear i el transport actiu d'ARNm neuronals al llarg dels microtúbuls. Com que un dels factors també funciona com a regulador de longitud de la regió no traduïda (3'UTR), un determinant recentment identificat de la localització de l'ARNm, aquesta tesi també estableix les bases per estudiar i entendre l'ordenació de l'ARNm depenent de la longitud de 3'UTR a les neurones.
