Abstract

The intrinsic complexity and dynamic nature of B-lymphocytes make them ideal for in-depth analysis using single-cell technologies. Secondary lymphoid organs (SLOs) are central in B-lymphocyte activation and differentiation, but a detailed understanding of their cellular networks and regulatory mechanisms is still lacking. This thesis utilizes human tonsils as a model of SLOs to generate a detailed census of tonsillar cell types and states, with special focus on the comprehensive characterization of B-lymphocyte differentiation pathways, highlighting their gene regulatory mechanisms and spatial determinants. Although naturally protective, dysregulation of B-lymphocytes activities can be harmful, yet a holistic understanding of their role in various inflammatory diseases remains elusive. This thesis also maps the immune plasticity across 18 inflammatory-diseases, revealing novel insights on B-lymphocytes and interferon-response genes in Systemic Lupus Erythematosus pathogenesis. Altogether, this work enriches our understanding of immune cell dynamics, shedding light on their multifaceted functions in health and disease and opening avenues for improved diagnosis and treatment strategies.
Los linfocitos-B son células heterogéneas por naturaleza, lo que las hace ideales para su análisis mediante tecnologías de célula única. A pesar de que los órganos linfoides secundarios (OLS) juegan un papel clave en su activación y diferenciación, todavía no existe un conocimiento detallado de sus redes celulares y mecanismos reguladores. Esta tesis estudia las amígdalas humanas, como modelo de OLS, para generar un censo detallado de sus tipos y estados celulares, centrándose en la caracterización exhaustiva del proceso de diferenciación de los linfocitos-B. Las actividades de los linfocitos-B son generalmente protectoras, pero su desregulación puede resultar perjudicial y ser crítica en la patogénesis de diferentes enfermedades. Por ello, esta tesis también estudia la plasticidad inmune en 18 enfermedades inflamatorias, revelando nuevos conocimientos sobre la relación de los linfocitos-B y los genes de respuesta al interferón en la patogénesis del lupus eritematoso sistémico. En conjunto, esta tesis aporta una comprensión holística sobre la dinámica de las células inmunitarias, esclareciendo sus funciones multifacéticas en pacientes sanos y enfermos, y posibilitando el desarrollo de nuevas estrategias de diagnóstico clínico y tratamiento.
