Abstract

Type 1 Diabetes (T1D) is an autoimmune disease marked by the selective destruction of pancreatic β cells. Early insulitis involves interactions between immune cells and β cells, but the mechanisms leading to β cell death are not fully understood. This thesis develops an in vitro co-culture model of β cells with CD4+ T cells to mimic early insulitis and study β cell responses. Cytokine profiling confirmed IFNγ presence and other inflammatory cytokines, indicating bidirectional communication. A significant chromatin remodeling and extensive gene expression changes were observed, identifying new regulatory elements responsive to the stimulus, termed Induced (IREs) and Lost (LoREs) Regulatory Elements. Some of the IREs overlap T1D-risk SNPs, variants that might disrupt normal β cell responses to inflammation. CRISPR activation experiments validated the functional impact of IREs on SOCS1 gene expression. The co-culture model provides insights into β cell responses to immune-mediated inflammation and helps identify potential therapeutic targets for T1D treatment.
La diabetis tipus 1 (T1D) és una malaltia autoimmune caracteritzada per la destrucció selectiva de les cèl·lules β pancreàtiques. La inflamació dels illots pancreàtics, insulitis, implica interaccions entre les cèl·lules immunitàries i les cèl·lules β. No obstant això, els mecanismes exactes que condueixen a la mort de les cèl·lules β no són del tot coneguts. En aquesta tesi es presenta un model in vitro de co-cultiu de cèl·lules β amb cèl·lules T CD4+ per imitar la insulitis i estudiar la resposta de les cèl·lules β. La caracterització dels components del medi va confirmar la presència d'IFNγ i altres citoquines inflamatòries, indicant una comunicació bidireccional entre les dues poblacions. També es va observar una remodelació significativa de la cromatina i grans canvis en l'expressió gènica, identificant nous elements reguladors, anomenats Elements Reguladors Induïts (IREs) i Elements Reguladors Perduts (LoREs). Alguns IREs es superposen amb SNPs de risc de T1D, variants genètiques que podrien alterar la resposta de les cèl·lules β a la inflamació. Experiments amb CRISPRa han validat l'impacte funcional d’un IRE que conté un SNP de risc en l'expressió del gen SOCS1. El nou model de co-cultiu aporta informació sobre les respostes de les cèl·lules β a la inflamació i pot ajudar en la identificació de noves dianes terapèutiques per al tractament de la T1D.
