Abstract

Eukaryotic DNA is condensed inside the nucleus, compacted into chromatin. The structure of chromatin is dynamic, and its accessibility to cellular machinery can be altered depending on the level of chromatin compaction (euchromatin or heterochromatin) and the induction of specific transcription programs. In Schizosaccharomyces pombe, the MAP kinase Sty1 and the transcription factor Atf1 play a key role in the regulation of gene expression during diverse biological processes. These inducible transcriptional responses are essential to survive to different environmental changes, and phosphorylated Atf1 rules the chromatin remodeling needed to adapt and survive. Furthermore, the transcription factor Atf1 does not only affect euchromatic transcription, but also the formation and maintenance of heterochromatin. In this work, I describe the role of Sty1-mediated phosphorylation of Atf1 in different processes, both stress related or not, and the effects caused in the chromatin dynamics by this pathway.
En eucariotas, el DNA se condensa en el núcleo en forma de cromatina. La estructura de dicha cromatina es dinámica, y que sea accesible o no a la maquinaria celular depende del nivel de compactación de la cromatina (eucromatina o heterocromatina) y de la inducción de programas transcripcionales concretos. En Schizosaccharomyces pombe, la MAP quinasa Sty1 juega un papel fundamental en la regulación de la transcripción durante múltiples procesos biológicos, especialmente a través del factor de transcripción Atf1. Esta respuesta transcripcional inducible es crucial para la supervivencia a distintos estreses, y necesita que Atf1 esté fosforilado para desencadenar los cambios en la cromatina necesarios para adaptarse y sobrevivir. Pero Atf1 no solo afecta a la transcripción de eucromatina, puesto que tambíen participa en la formación y mantenimiento de heterocromatina. En esta tesis doctoral, describo el papel de la fosforilación de Atf1 causada por Sty1 en diversos procesos, sean relacionados con estrés o no, y los efectos que causan en la cromatina.
