Abstract

The exocyst is an heterooctameric protein complex that tethers secretory vesicles to the plasma membrane during exocytosis. Each exocytic event is mediated by 10-15 exocysts. However, how multiple exocysts organize in time and space to accomplish their function remains unknown. We resolved the dynamic higher-order structure of the exocyst by correlating live-cell imaging and super-resolution microscopy. Fluorescent temporal markers demonstrated that multiple exocysts follow dynamic higher-order structures that switch from small and compact clusters to wider ring-like organizations with an asymmetric distribution of the exocysts around a central cavity. Conditional mutants linked conformational changes with defined molecular functions and cryo-electron tomography provided accurate descriptors of the vesicle geometry. We propose a model were the multiple exocysts bind the vesicle as compact clusters for four seconds. In a Sec9-dependent process, the exocysts open to form stable arcs and ring-like architectures that are disassembled by Sec18 eight seconds after tethering initiation.
L’exocyst és un complex proteic que uneix vesícules de secreció a la membrana plasmàtica durant l’exocitosi. Entre 10 i 15 exocysts participen en cada esdeveniment d’exocitosi, però en desconeixem la seva l’organització espacial i temporal. La combinació de microscòpia en cèl·lules vives i de super-resolució ens ha permès resoldre la supra-estructura de l’exocyst. Mitjançant l’ús de marcadors temporals hem demostrat que aquesta supra-estructura és dinàmica: comença formant clústers compactes que es transformen en estructures anulars al voltant de les quals els exocysts es distribueixen asimètricament. Gràcies als mutants condicionals hem assignat canvis conformacionals a funcions moleculars concretes, i la crio-tomografia electrònica ens ha permès descriure geomètricament les vesícules. Proposem un model on els exocysts s’uneixen a la vesícula formant clústers durant 4 segons. Sec9 indueix l’obertura d’aquests exocysts, que passen a formar arcs i anells estables. 8 segons després de l’inici del procés, l’acció de Sec18 desmunta aquests anells.
