Abstract

The activation of Notch1 receptor triggers differential outcomes depending on the upstream activating ligand. This way, Jag1-Notch1 activation promotes endothelial to hematopoietic transition process for the generation of hematopoietic stem cells, whereas Dll4-Notch1 activation promotes vasculo-arterial specification. Nevertheless, the mechanistics that selectively induce one fate or the other have not been characterized so far. Therefore, this thesis work aimed to uncover the particulars of how Notch pathway orchestrates HSC specification in the aorta-gonad-mesonephros region. We have shed light on the particular effects triggered by Jag1 and Dll4 ligands and we propose a cross-talk regulation between the two ligands as the mechanism by which Notch acquires the right signal strength and exerts the decision between arterio-endothelial and hematopoietic fate specification.
L’activació del receptor Notch1 comporta resultats diferencials que depenen del lligand que l’activi a un nivell superior de la via. D’aquesta manera, l’activació Jag1-Notch1 promou el procés de transició endoteli-hematopoètica per la generació de cèl•lules mare hematopoètiques, mentre que l’activació Dll4-Notch1 promou l’especificació vasculo-arterial. No obstant això, els mecanismes que indueixen selectivament un destí o altre no han estat caracteritzats. Per aquest motiu, aquesta tesi té per objectiu caracteritzar com la via de Notch dirigeix l’especificació de cèl•lules mare en la regió de l’aorta-gonada-mesonefros. En aquest treball hem caracteritzat els efectes específics activats pels lligands Jag1 i Dll4 i proposem una regulació creuada entre aquest dos lligands com a mecanisme per mitjà del qual Notch assoleix el nivell d’activació requerit per decidir entre especificació arterio-endotelial i destí hematopoètic.
