Abstract

Zinc is an essential micronutrient involved in many different biological processes. Zinc homeostasis is mainly regulated by 24 zinc transporters coded by the Zinc Transporter Genes (ZTGs). Notably, zinc body content is highly dependent on environmental zinc availability, and its deficiency is life-threatening for several human populations. Since differences in zinc homeostasis may be adaptive to local dietary and environmental conditions, we explored the complete set of 24 human ZTGs for signatures of positive selection using genomes from worldwide populations in the 1000 Genomes Project and South Asian populations in the 100K GenomeAsia Project. We have identified unusual patterns of variation resulting from strong selective sweeps on a few ZTGs but also from polygenic selection on the whole set of ZTGs, especially in South Asia. To understand the evolutionary history of the strong signal of positive selection we confirmed in SLC30A9 and fill the gap between genotype and phenotype, we also explored the functional implications of a putative candidate variant behind this case of adaptation.
El zinc és un micronutrient essencial implicat en múltiples processos biològics. L’homeòstasi del zinc està principalment regulada per 24 transportadors de zinc codificats pels Zinc Transporter Genes (ZTGs). El contingut de zinc a l’organisme depèn de la disponibilitat de zinc ambiental, i el seu dèficit és una causa de malaltia greu en diverses poblacions humanes. Atès que les diferències en l’homeòstasi del zinc podrien ser el resultat de l’adaptació a condicions locals, hem explorat el conjunt dels 24 ZTG humans buscant senyals de selecció positiva en genomes de poblacions d’arreu del món del Projecte 1000 Genomes i poblacions del sud d’Àsia del Projecte GenomeAsia 100K. Hem identificat patrons de variació inusuals que resulten de forts escombrats selectius en alguns ZTG, però també de la selecció poligènica en tot el conjunt de ZTG, especialment al sud d'Àsia. Per entendre la història evolutiva del fort senyal de selecció positiva que hem confirmat al gen SLC30A9 i omplir el buit entre el seu genotip i fenotip, hem explorat les implicacions funcionals d'una variant candidata per aquest cas d’adaptació.
