Abstract

Temperature affects all processes, including development, and yet, it remains unknown how and/or if underlying mechanical and biochemical events are coordinated to slow down development robustly at lower temperatures. Here, using a variety of methods from molecular biology to 5D live imaging, I uncover the vulnerabilities of the zebrafish epithelial during gastrulation at sub-optimal temperatures (20° C). The enveloping layer (EVL) in the cold displays large multi-nucleated cells stemming from cytokinesis failures. Differences in tissue tension during late epiboly also led to rupturing of the EVL in certain embryos. Furthermore, mesendoderm patterning reveals convergence-extension defects under varying temperatures, suggesting that different aspects of gastrulation are affected in parallel. Proteome profiling across five different temperatures also reveals processes affected under temperature fluctuations. This PhD project is one of the first studies focusing on the effect of temperature on gastrulation and exposes mechanisms behind developmental failures under sub-optimal temperatures.
La temperatura afecta a tots els processos i, no obstant això, es desconeix com i/o si els esdeveniments mecànics i bioquímics es coordinen per alentir el desenvolupament a temperatures més baixes. Aquí, utilitzant des de la biologia molecular fins a la microscòpia en 5D, descobrim les vulnerabilitats de la gastrulació en l'epiteli del peix zebra a temperatures subòptimes (20° C). La capa envoltant (EVL) en fred mostra cèl·lules multinucleades derivades de fallades de citocinesi. Diferències en la tensió tissular durant l'epibòlia tardana també provoquen la ruptura de l'EVL en certs embrions. A més, el patró de mesoderma revela defectes d'extensió-convergència sota temperatures variables, suggerint que diferents aspectes de la gastrulació es veuen afectats. L'anàlisi proteòmic en cinc temperatures diferents també revela processos afectats per les fluctuacions de temperatura. Aquest projecte de doctorat és dels primers en centrar-se en l'efecte de la temperatura sobre la gastrulació i exposa els mecanismes rere conseqüents fallades de desenvolupament.
