Abstract

Neural microexons are the most conserved type of alternative splicing, suggesting a strong functional relevance. Consistently, mutation of their master regulators leads to dramatic neurodevelopmental alterations and autistic-like behaviors. However, the functional relevance of individual microexons has only been elucidated for a few cases. Here, I aimed at investigating the roles of individual microexons using zebrafish as a model system. We selected 18 microexons conserved from fish to humans and generated deletion lines using CRISPR-Cas9. None of these deletions produced gross phenotypic alterations or affected viability and fertility. However, a finer characterization using different tests I have developed or adapted during my PhD revealed a wide range of phenotypes at the cellular, tissular, behavioral and/or molecular level. Interestingly, in many cases, these phenotypes were in opposite directions, a pattern that was mirrored at the transcriptomic level. Altogether, our results depict a rich scenario for the functionality of microexons.
Els microexons neurals són el tipus més conservat de “splicing” alternatiu, fet que indica una gran rellevància funcional. A més a més, s’ha observat que mutacions produïdes en els seus principals reguladors desencadenen conseqüències dràstiques en el neuro-desenvolupament i també produeixen fenotips que s’aproximen a comportaments observats en l’autisme. Tot i així, les funcions concretes dels microexons individuals han estat descrites en pocs casos. L’objectiu principal d’aquest projecte és investigar les funcions individuals dels microexons. Per dur-ho a terme hem utilitzat el peix zebra com a model animal. Hem seleccionat 18 microexons, conservats entre peixos i humans, i hem generat línies mutants de CRSPR-cas9 per deleccionarlos. Cap de les línies generades mostra alteracions fenotípiques rellevants ni presenta alteracions en la viabilitat o fertilitat dels individus. Tanmateix, mitjançant una caracterització acurada dels fenotips i emprant diferents tests, que he desenvolupat i adaptat durant el transcurs del doctorat, hem detectat tot un ventall de fenotips a nivell cel·lular, tissular, molecular i conductual. Curiosament, en la majoria dels casos els fenotips van en direccions oposades, un patró que veiem perfectament reflectit a nivell transcriptòmic. Els resultats en conjunt, ens mostren un escenari molt ric per la funcionalitat dels microexons
