Abstract

Migraine is one of most disabling neurological disorders for which the genetic and molecular mechanisms are not fully resolved. This PhD thesis has focused on the functional characterization of two new missense mutations in CACNA1A, the pore forming subunit of the CaV2.1 channel, associated to migraine. The analysis of these mutations revealed that the functional interactions of CaV2.1 mutant channel with regulatory proteins implicated in synaptic transmission (i.e. G proteins and SNARE proteins) are altered and may be responsible for the observed clinical phenotype. Moreover, our studies also propose that mutations in CACNA1A can be not only causative of the disease but modifiers of the migraine symptoms as well.
La migranya és un dels trastorns neurològics més incapacitant les causes genètiques i moleculars del qual no estan del tot resoltes. Aquesta tesi doctoral s’ha centrat en la caracterització funcional de dues noves mutacions amb canvi de sentit en CACNA1A, la subunitat formadora del porus del canal CaV2.1, associades a migranya. L’anàlisi d’aquestes mutacions ha desvetllat que les interaccions funcionals del canal mutant CaV2.1 amb proteïnes reguladores implicades en la neurotransmissió (proteïnes G i proteïnes SNARE) estan alterades i poden ser responsables dels fenotips clínics observats. A més, els nostres estudis proposen que mutacions en CACNA1A poden no només ser causa de la malaltia sinó també modificar-ne els símptomes.
