Abstract

El conegut supressor tumoral p53 regula un ampli programa transcripcional que protegeix envers el càncer. No obstant, la complexitat d’aquest programa fa difícil determinar el rol de cada gen regulat per p53. Deplecionar RNF144B, una ligasa d’ubiquitina E3 regulada per p53, incrementa el desenvolupament i severitat de tumors de linfoma en ratolins. Aquí, estudio si RNF144B també és un supressor tumoral fora del compartiment sanguini. Mitjançant transcriptòmica, proteòmica, genòmica y anàlisis funcional de cèl·lules humanes i murines amb funció normal de p53, revel·lo un rol de RNF144B en la proliferació i transformació cel·lular, la reparació del ADN i el manteniment de l’estabilitat genòmica. En el context dels adenocarcinomes pulmonars, la deficiència de RNF144B porta a la resistència enfront certs fàrmacs citotòxics, una pitjor supervivència a la malaltia dels pacients. Conjuntament, aquestes troballes situen RNF144B com un supresor tumoral regulat per p53 en altres contextes cel·lulars com el càncer de pulmó.
The well-known tumor suppressor protein p53 regulates a vast transcriptional program that protects against cancer development. However, given the complexity of this network it is difficult to understand the role of each of the p53 targets. Knockdown of RNF144B, a p53-regulated E3-ubiquitin ligase, enhanced the rate of tumour development and severity in murine Eu-MYC lymphomas. In this work I aimed to determine if RNF144B has an impact on tumor suppression beyond the hematopoietic compartment. Through integrative transcriptomics, proteomics, genomics and functional analysis of wild type p53 human and mouse cells I revealed a role for RNF144B in coordinating cellular proliferation and transformation, DNA repair and genome stability. In the context of lung adenocarcinomas, RNF144B deficiency leads to resistance to cytotoxic drugs, and patients have a worse disease-survival overall. Altogether, my findings indicate that RNF144B acts as p53-activated tumor suppressor in other cellular contexts, especially in the lung compartment.
