Abstract

The outer epithelial layer is the first cell type that emerges during development, providing essential mechanical protection and is indispensable for morphogenesis. Here, we study how the interplay between molecular and mechanosensing pathways participate in this tissue formation and function by combining in vivo assays with bottom-up ex vivo/in vitro assays using zebrafish enveloping layer (EVL) as model system. We found that mechanosensitive signalling pathways involving Trpv4 (transient receptor potential cation channel subfamily V member 4) activity at the plasma membrane and cPLA2 (cytoplasmic phospholipase A2) activity at the inner nuclear membrane, together with the transcription factors CCAAT/enhancer-binding protein-α (C/EBPα) and -β (C/EBPβ), are involved in regulating EVL cellular/nuclear shape, contractility and specification, thus affecting early development when perturbed. Moreover, we established ex vivo and in vitro systems that are able to recapitulate de novo EVL formation.
La capa epitelial externa és el primer tipus cel·lular que emergeix durant el desenvolupament, proporcionant una protecció mecànica essencial i sent indispensable per a la morfogènesi. En aquest estudi, investiguem com la interacció entre vies moleculars i mecano-sensitius participa en la formació i funció d'aquest teixit mitjançant l'ús combinat de proves in vivo i ex vivo/in vitro utilitzant l’enveloping layer (EVL) del peix zebra com a sistema model. Hem descobert que les vies de senyalització mecano-sensibles que involucren l'activitat del Trpv4 (canal catiónic de potencial receptor transitori de la subfamília V) a la membrana plasmàtica i l'activitat del cPLA2 (fosfolipasa A2 citoplasmàtica) a la membrana nuclear interna, juntament amb els factors de transcripció CCAAT/enhancer-binding protein-α (C/EBPα) i -β (C/EBPβ), estan implicats en la regulació de la forma cel·lular/nuclear, contractilitat i especificació de l'EVL, afectant així el desenvolupament primerenc quan es pertorben. A més, hem establert sistemes ex vivo i in vitro que són capaços de recapitular la formació de l'EVL de novo.
