Abstract

Cells commonly live in a crowded environment, in a tissue or in a developing embryo. The extracellular matrix and/or surrounding neighboring cells can impose physical deformations to which cells need to adapt in order to efficiently migrate, proliferate or differentiate and to build functional 3D tissues. Here we show that the nucleus allows cells to measure mechanical shape deformations and regulates cell contractility by the activation of a mechanosensitive signaling pathway at the inner nuclear membrane, leading to the transformation into a motile stable-polarized amoeboid phenotype. We found that upon cell polarization, nuclei drastically change their shape and deform and acquire a nuclear polarization that aligns with the cell polarization axis. This deformation is dependent on the actomyosin cortex, nuclear rigidity and the connection between the nucleus and cytoskeleton elements. All together, our data support a close mechanical cross-talk between cell shape deformation and nucleus architecture, involved in the mechanosensitive regulation of cell plasticity and adaptive cell behaviour in 3D tissues.
Ya sea en un tejido o durante el desarrollo embrionario, las células están comprendidas en un entorno superpoblado. La matriz extracelular y/o las células vecinas pueden imponer deformaciones físicas a las que las células necesitan adaptarse para poder migrar eficientemente, proliferar o diferenciarse para construir tejidos 3D funcionales. Aquí demostramos que el núcleo celular, por un lado permite a la célula medir las deformaciones mecánicas y, por el otro regular la contractilidad celular atravesó de vía de señalización mecano sensible, situada en la membrana nuclear interna. Estas señales confieren a la célula un fenotipo migratorio llamado polarización-estable amoeboide. Demostramos que tras la polarización celular, el núcleo cambia drásticamente su forma y adquiere una polarización nuclear que se alinea con el eje de polarización celular. Esta deformación depende del córtex de actomiosina, la rigidez nuclear y la conexión entre el núcleo y los elementos del citoesqueleto. En conjunto, nuestros datos respaldan una estrecha relación mecánica entre la deformación de la forma celular y la arquitectura del núcleo, involucrada en la regulación mecanosensible de la plasticidad celular y el comportamiento adaptativo en tejidos 3D.
