Abstract

The molecular mechanisms regulating sperm differentiation (spermiogenesis) and maturation in fishes are largely unknown. In these oviparous species, the initiation of sperm motility is triggered by the osmolality of the aquatic environment into which they are ejaculated. However, the regulatory signals involved in the acquisition of the spermatozoon full motility potential, as well as the molecular factors required for the maintenance of the spermatozoon swimming performance remain unclear. This thesis investigated the transcriptomic and protein networks during spermiogenesis and sperm maturation in the marine teleost gilthead seabream (Sparus aurata) and examined the role of ion channels in the prolongation of the motility of post activated spermatozoa in the hyperosmotic seawater. The findings uncover new signaling pathways regulating sperm maturation and show that spermatozoon cytoplasmic translation is important during this process. In addition, the role of some K+- and Ca2+-selective channels, and of other stretch-activated and mechanosensitive channels, in the control of the spermatozoon swimming pattern was identified. Finally, the investigations revealed that some of the Ca2+-permeable channels interact with aquaporin-4a to trigger regulatory volume increase mechanisms that maintain spermatozoon viability and motility under hypertonic conditions.
Els mecanismes moleculars que regulen la diferenciació dels espermatozoides (espermiogènesi) i la maduració en els peixos són en gran part desconeguts. En aquestes espècies ovípares, l'inici de la motilitat dels espermatozoides es desencadena per l'osmolalitat del medi aquàtic en el qual s'ejaculen. No obstant això, els senyals reguladors implicats en l'adquisició del potencial de motilitat dels espermatozoides, així com els factors moleculars necessaris per al manteniment de la natació encara no estan clars. En aquesta tesi, s’han investigat les xarxes transcriptòmiques i proteòmiques durant l'espemiogènesi i la maduració dels espermatozoides a l'orada (Sparus aurata), i s’ha examinat el paper dels canals iònics en la prolongació de la motilitat dels espermatozoides post activats en l'aigua de mar. Les troballes descobreixen noves vies de senyalització que regulen la maduració dels espermatozoides i mostren que la traducció citoplasmàtica és important durant aquest procés. A més, s’ha identificat el paper d'alguns canals de K+ i Ca2+, i d'altres canals activats per estirament i mecanosensibles, en el control del patró de natació dels espermatozoides. Finalment, les investigacions han revelat que alguns dels canals de Ca2+ interactuen amb l'aquaporina-4a per desencadenar mecanismes d'increment del volum regulador que mantenen la viabilitat i la motilitat dels espermatozoides en condicions hipertòniques.
