Abstract

Ageing is a very complex phenotype that has been widely studied, but is still largely unknown. Most of the studies focus on a single species, or on performing genome wide association studies in human cohorts, but this only provides a glimpse into the genetics that shaped the lifespan of a species, since some variants are fixed or segregate at very low frequency in a given species, and thus cannot be detected by these methods. Instead, we studied ageing from an evolutionary perspective; first, by using comparative genomics, taking advantage of its phenotypic diversity in mammals to gain insight into which genes and pathways have contributed to its diversity; and second, by studying the shared genetic architecture -pleiotropy- between early and late onset diseases and, as a detailed case study, between longevity and two early onset diseases with a relatively high prevalence, schizophrenia and bipolar disorder, to discover new loci influencing human longevity and to test evolutionary theories of ageing.
L'envelliment és un fenotip molt complex i àmpliament estudiat, però encara està ple d'incògnites. La major part dels estudis se centren en una única espècie, o en realitzar estudis d'associació del genoma complet en cohorts humanes, però això només ofereix un cop d'ull a la genètica que ha contribuït a establir la longevitat d'una determinada espècie, ja que algunes variants hi estan fixades, o apareixen en molt baixa freqüència, impedint la seva detecció amb aquests mètodes. En canvi, nosaltres vam estudiar l'envelliment des de la perspectiva de l'evolució; primer, mitjançant la genòmica comparativa, aprofitant la seva diversitat fenotípica en mamífers per comprendre millor quins gens i rutes hi han contribuït; i segon, estudiant la pleiotropia entre malalties d'aparició precoç o tardana i, com a estudi detallat, entre la longevitat i dues malalties d'aparició precoç amb prevalença relativament alta, esquizofrènia i trastorn bipolar, per a descobrir nous loci que afecten la longevitat humana i per a avaluar les teories evolutives de l'envelliment.
