Abstract

Embryonic stem cells (ESCs) are derived from the inner cell mass of the pre-implantation embryo and can recapitulate early developmental stages of the embryo. ESCs have the ability to continuously propagate while maintaining the capacity to differentiate into all cell lineages of the epiblast. The Wnt/ β-catenin signalling pathway plays a fundamental role in controlling the mouse ESC regulatory circuit. However, the downstream targets of the Wnt pathway contributing to the process of pluripotency remain unknown. Here we used computational analysis to identify Gli2 as a putative downstream target of Wnt, a regulator of mESC pluripotency and a potential regulator of early embryo development. Investigating the role of this factor gave us further insight into ESC and embryo biology.
Les cèl·lules mare embrionàries (ESC) es deriven de la massa cel·lular interna de la preimplantació embrionària i poden recapitular les primeres etapes del desenvolupament embrionari. Els ESC tenen la capacitat de propagar-se contínuament, mantenint la capacitat de diferenciar-se en tots els llinatges cel·lulars de l'epiblast. La via de senyalització Wnt / β-catenin té un paper fonamental en el control del circuit regulador ESC del ratolí, tanmateix, els objectius de la via Wnt que contribueixen al procés de pluripotència encara són desconeguts. Aquí, mitjançant l’anàlisi computacional, vam identificar un objectiu putatiu Wnt Gli2 com un regulador de la pluripotència de mESC i un regulador potencial en el desenvolupament primerenc d'embrions. Investigar el paper d'aquest factor ens va donar una visió de l'ESC i la biologia de l'embrió.
